BSTR酶反应器中布洛芬的酶促拆分及反应动力学研究Resolution of Racemic Ibuprofen in BSTR Enzymatic Reactor and Kinetic Study of the Reaction

摘要(Abstract):
为了提高固定化扩展青霉TS414脂肪酶(PEL)对外消旋布洛芬的拆分效率,利用均匀设计的实验方法,在酶反应器中考察和优化了布洛芬、固定化脂肪酶、1-丙醇等几个参数对拆分反应的影响.优化后的实验结果表明:当反应体系中布洛芬为200 mg,固定化酶的加量为6 g,1-丙醇为0.383 mL,反应达到平衡时,布洛芬拆分反应的底物转化率接近50%,底物的对映体过量值达到94.3%,产物的对映体过量值则稳定在98%以上.而且固定化PEL催化拆分反应的周期缩短至12 h.同时,基于乒乓反应机制的动力学方程,建立了相应的拆分反应动力学模型,并得到了该模型的有关参数.
