黑脊倒刺鲃scp3基因的克隆及其表达Molecular Cloning and Expression of scp3 Gene in Spinibarbus caldwelli

摘要(Abstract):
为了从分子水平上研究鱼类生殖细胞发育的机制,首次从重要的经济养殖鱼类黑脊倒刺鲃中克隆出了生殖细胞减数分裂Ⅰ相关基因scp3的部分同源序列scscp3:该序列长477bp,编码159个氨基酸.经比对发现,其编码的蛋白序列与斑马鱼scp3蛋白等具有高度的同源性.不同组织RT-PCR结果显示:scscp3基因仅在生殖细胞中表达,可将其作为生殖细胞的标记基因,用于鱼类生殖发育的研究.RNA原位杂交结果表明:在精子发生中,scscp3基因均是在精原细胞和初级精母细胞中表达,在后期的精子细胞中未发现表达.而在卵子发生中,scscp3基因在卵原细胞和卵母细胞的各个时相中均有表达,并且在卵原细胞和前期的卵母细胞中表达强烈,在后期时相的卵母细胞中阳性信号逐渐减弱,且向细胞边缘聚集.
