一株具抗肿瘤活性的红树林细菌筛选及成分分离Isolation of an Anti-tumor Bacteria and Extraction of Anti-tumor Substances

摘要(Abstract):
为从红树林微生物中分离抗肿瘤活性物质,从泉州湾红树土壤中分离、纯化和筛选能产生抗肿瘤活性物质的细菌,并利用16S rDNA序列比对方法进行分子鉴定.采用追踪分离的方式,以稻瘟霉分生孢子抑制强度和肿瘤细胞体外抑制活性为指标,对细菌发酵液抗肿瘤活性成分进行初步分离.实验结果显示,从泉州湾红树林土壤中分离出1株具有较强抗肿瘤活性的细菌,经鉴定为Vibrio fluvialis. Vibrio fluvialis发酵液能抑制稻瘟霉分生孢子萌发时菌丝形变,并抑制EC9706食管癌细胞和H22肝癌细胞生长.从Vibrio fluvialis发酵液分离出具有较强抗肿瘤活性的化学组分Ⅰ和组分Ⅱ.组分Ⅰ对EC9706食管癌细胞和H22肝癌细胞ρIC50分别为0. 18 mg·m L(-1)和0. 26 mg·m L(-1)和0. 26 mg·m L(-1),组分Ⅱ对EC9706食管癌细胞和H22肝癌细胞ρIC50分别为0. 17 mg·m L(-1),组分Ⅱ对EC9706食管癌细胞和H22肝癌细胞ρIC50分别为0. 17 mg·m L(-1)和0. 22 mg·m L(-1)和0. 22 mg·m L(-1).表明Vibrio fluvialis能产生抗肿瘤活性物质,具有重要抗肿瘤研发价值.
