重组工程菌E.coli BL21(DE3)-pET29a-aiiA可溶性AiiA蛋白的发酵条件优化Optimization of the Fermentation Conditions of Recombination E.coli BL21(DE3)-pET29a-aiiA for the Expression of Soluble AiiA Protein

摘要(Abstract):
通过对工程菌E.coli BL21(DE3)-pET29a-aiiA的摇瓶发酵培养条件进行优化,确定了AiiA蛋白可溶性表达的最佳发酵培养基为牛肉膏蛋白胨.目的蛋白可溶性表达的最佳诱导条件为:IPTG浓度0.6 mmol/L,诱导初始OD600为0.5,诱导时间为20 h,温度20℃,pH为7.0,微量元素Na2HPO420 mmol/L,250 mL三角瓶的装液量为70 mL.优化前可溶性AiiA蛋白的相对含量为2.23 mg/mL,占总目的蛋白的31.4%,优化后达到4.54 mg/mL,占总目的蛋白的60.3%,大大提高了目的蛋白的可溶性表达.
