恒河猴牙髓干细胞的牙向分化Tooth Differentiation of Rhesus macaques Dental Pulp Stem Cells

摘要(Abstract):
将恒河猴牙髓剪碎,用酶消化后培养获得恒河猴牙髓干细胞,用细胞免疫荧光的方法鉴定干细胞表面标记基因的表达,对分离的细胞进行成脂成骨的诱导,同时将细胞与支架混合后移植裸鼠皮下.结果发现,分离的细胞能持续传代,能被条件性地诱导成脂和成骨,能表达CD146,OCT4,SSEA4,STRO1等干细胞的标记基因,细胞与支架移植体内能分泌形成牙本质.结果表明酶消化法能够成功分离恒河猴的牙髓干细胞,该细胞在体内能分化形成牙本质.实验结果为该细胞进一步作为种子细胞用于牙齿再生研究奠定了实验基础.
